Lupine Lane is a short street in Ranlett Grove, South Billerica, running south from Ossamequin Road across Osceola Lane. It was first laid out on paper on the plan "Ranlett Grove on shore of Concord River, Billerica Mass.," drawn in May 1909 by Ernest W. Branch, civil engineer, for Atwood and Pattee, auctioneers and real estate dealers of 27 School Street, Boston, and recorded on Aug. 7, 1909 (Plan Book 26, Plan 47). On the plan the lane leaves Ossamequin Road between lots 16 and 17, crosses Osceola Lane and continues south past lots 103 to 106. The grove was named for the estate of Captain Charles A. Ranlett, the sea captain who had lived on the Bedford road from 1862 to 1878.

The lane is named for the lupine, the wildflower. The plan does not say so, but it belongs to a group of flower and bird names on the same plan (Laurel Lane, Cardinal Lane, Robin's Lane) set beside native names such as Ossamequin and Osceola.

Lupine Lane was accepted as a public way at the annual town meeting on May 13, 1976, under Article 37, the year's long list of street acceptances, where the warrant printed it as "Lupine Road." The motion was "to accept the laying out of LUPINE LANE in accordance with a plan on file with the Town Clerk a way 50 feet wide and 743 feet long"; an amendment to raise $235 for repairs was lost, and the acceptance carried. The Ranlett Grove area was sewered in 1995 and 1996. The six inch water main in the lane, which had failed eleven times, was replaced in August 2015 with 1,224 feet of eight inch ductile iron pipe. Lupine Lane is not among the roads of the 1831 town map accepted by town meeting in October 2016.
